Hantavirus Infection of SD Rats Reared in Domestic Laboratory Animal Facilities

Abstract
IFA (Indirect Fluorescence Antibody) method and Western blotting analysis were applied as serological diagnosis to SD rats reared in the domestic animal ficilities. Vero cells infected with Seoul virus strain SR-11 and recombinant Hantaan 76-118 neucleoprotein expressed in baculovirus were used as antigens in IFA method and Western blotting analysis, respectively. When IFA method was applied to total B06 rats sera diluted 1:32, strong positive reaction was observed in 0.66%, positive reaction was observed in 11.55% and positively suspected reaction was observed in 16.34% of the rats. Western blotting analysis was applied to the rats sera diluted 1:32 which showed strong positive and positive reaction in IFA method. Among total 74 rats sera, only 8 sera showed 25kDa, 30kDa and 48kDa bands, meaning positive reaction in Western blotting analysis. The positive reaction in Western blot analysis was found in the rats reared in conventional animal ficilities.
In this study, we confirmed that double serological check was required for diagnosis hantavirus infection in laboratory rats.
